  • 소개 및 적용: Neratinib (HKI-272) is an oral, irreversible dual inhibitor of EGFR and HER2, which are receptors known to play a central role in cell proliferation and cancer development. It is highly selective and has potent antitumor activity against breast and non-small cell lung cancer. Neratinib has an IC50 of 59 nM for HER2 kinase and 92 nM for EGFR, and shows greater selectivity over other kinases. As an irreversible inhibitor, it may overcome resistance to reversible inhibitors like gefitinib. Neratinib has been evaluated in clinical trials for cancers with HER2 overexpression or activating mutations.
